New insights into regulation of p53 protein degradation

چکیده
The tumor suppressor p53 is a multi-functional protein, its functions covering cell cycle control, apoptosis, senescence, genome integrity maintenance, metabolism, cellular reprogramming and autophagy. Regulation of p53 stability is a central process in controlling these p53 functions. Multiple E3 ligases that mediate p53 degradation through the ubiquitination proteasome pathway have been well studied, studies also suggest p53 can be degraded through the ubiquitination-independent pathway and the proteasome-independent pathway. This review discusses these mechanisms involved in p53 protein degradation aims at establishing a better understanding on regulation of p53 protein.

The Role of Tumor Protein 53 Mutations in Common Human Cancers and Targeting the Murine Double Minute 2–P53 Interaction for Cancer Therapy
The gene TP53 (also known as protein 53 or tumor protein 53), encoding transcription factor P53, is mutated or deleted in half of human cancers, demonstrating the crucial role of P53 in tumor suppression. There are reports of nearly 250 independent germ line TP53 mutations in over 100 publications. The P53 protein has the structure of a transcription factor and, is made up of several domains. T...
متن کاملالقای آپوپتوز وابسته به p53 در ردهی سلولی لوسمی لنفوبلاستیک حاد پیشساز لنفوسیت B (NALM-6) توسط مولکول کوچک RITA
Background and Objective: The use of low-molecular-weight, nonpeptidic molecules that degrade the interaction between the p53 protein and its negative regulator MDM2 (Murine- double minute colon 2) is a new therapeutic strategy for treatment of various types of cancer. One of these agents is RITA (reactivation of p53 and induction of tumor cell apoptosis) which binds to p53 protein and inhibits...

Fasting has numerous biological, physical and mental health advantages or that as some physicians cure their patients by prescribing fasting to them. Fasting protects people from many diseases such as cancer, cardiovascular diseases, and diabetes complications. The main health-promoting effects of fasting are increased production of neurotrophic factors, neuroendocrine activation, hormetic stre...
متن کاملHdmX stimulates Hdm2-mediated ubiquitination and degradation of p53.
The RING finger proteins HdmX and Hdm2 share significant structural and functional similarity. Hdm2 is a member of the RING finger family of ubiquitin-protein ligases E3 and targets the tumor suppressor protein p53 for degradation. Although HdmX also binds to p53, HdmX does not induce p53 degradation. Moreover, HdmX has been reported to interfere with p53 degradation in overexpression experimen...
متن کاملP-169: Protective Effect of Vitamin E on Cypermethrin - Induced Damages Correlates with P53 Gene Expression and Nitrosative Stress
Background: Because of fast rate of degradation and low mammalian cells toxicity the cypermethrin (CPM) is a wide used insecticide in domestic agriculture and in veterinary medicine applications. The compound exerts its pathological impact by down-regulating the antioxidant status. Therefore, current study was designed to evaluate the protective effect of vitamin E on CPMinduced apoptosis and n...
